Transcriptional Regulatory Mechanisms in Animal Cells

Robert Roeder’s career as a pioneer in the field of transcription began with his discovery of three forms of eukaryotic RNA polymerase in 1969, and led to some of the most fundamental findings within the field. He further showed that individual polymerases are specific for different classes of genes and require distinct sets of general initiation factors for promoter recognition and transcription. These findings were extended by identification of the first gene-specific transcription factor in eukaryotes and, more recently, a variety of transcriptional co-activators. These are currently studied, in conjunction with histone modifying cofactors, in defined cell-free systems reconstituted with recombinant chromatin templates.

“Reading the genome - from DNA sequence to expression”

Are complex transcriptional behaviors encoded in the DNA sequences of gene regulatory regions? Eran Segal claims ‘yes’ and he will present a quantitative computational framework to prove it. Based on the hypothesis of competitive binding equilibrium, he shows that this model explains many experimental observations regarding binding patterns of factors and nucleosomes, and dynamics of transcriptional activation.
